A strain-promoted 1,3-dipolar cycloaddition of a cyclooctynes and an azide was carried out forming a covalent linkage (also termed the Cu-free azide-alkyne cycloaddition). Thus, the aldehyde bearing proteins react under oxime formation with different heterobifunctional linkers which carry an aminooxy residue on one end and either an azide or cyclooctynes on the other.
